۱Development of New 96–Microwell–Based Spectrophotometric Assay with High–Throughput and its Application in Pharmaceutical Quality Control of Varenicline

This study describes, for the first, the development of a new 96–micorwell based spectrophotometric assay with high–throughput for the pharmaceutical quality control of varenicline (VRC). The reaction between VRC and 1,2–naphthoquinone–4–sulphonate (NQS) as a chromogenic reagent was investigated. In alkaline medium, a red–colored product exhibiting maximum absorption peak at 490 nm was produced. This colordeveloping reaction was employed, for the first time, in the development of the proposed assay. The reaction was carried out in 96–microwell plate and the absorbance of the colored–product was measured by microwell plate absorbance reader at 490 nm. The optimized reaction conditions were established; under which, Beer’s law was obeyed in the range of 10 – 100 μg ml 1of VRC. The limits of detection and quantification were 3.1 and 10.2 μg ml 1, respectively. The assay showed high precision as the values of relative standard deviations (RSD) did not exceed 2%. The proposed assay was applied successfully for the determination of VRC in its pharmaceutical tablets with good accuracy and precisions; the label claim percentages were 100.80 ± 1.62%. The proposed assay is practical and valuable in terms of its routine application in pharmaceutical quality control laboratories.
