Melissa officinalis L.due to its useful application in medicine, is being paid more attention. In order to establish a stable regeneration system with 4 landraces collected from different climate in Iran, major parameters such as cells fresh weight, cells dry weight, Packed Cell Volume, cells viability in the control and under influence of different sucrose levels were investigated. Statistical analysis of results showed sucrose level had significant impacts on the cells growth, so that increasing sucrose caused a significant increase in fresh weight. Among sucrose treatments the highest influence was achieved by 60 and 80 g\l treatments and the highest cells fresh weight was recorded in 10th day (80.6g) and 8th day (82.6g) respectively for 60 and 80 g\l treatments. The results obtained from cells dry weight showed the highest amount was recorded at 80 g\l sucrose at the 8th day (233.3mg\L). The comparative results due packed cell volume showed the highest amount of PCV for the sucrose treatment was recorded for 60 and 80 g\l.<\div>
