The purpose of present research was to identify a potential mixed culture obtained from a hot spring to oxidize ferrous iron Fe2+ to ferric iron Fe3+ used for the biodesulfurization process. This biological oxidation was carried out by several species of bacteria which has potential to oxidize ferrous irons the formed oxidizing agent was able to remove hydrogen sulfide from sour gases. the mixed culture was grown under aerobic condition . batch culture was conducted in an incubator–shaker at ambient temperature with a rotation speed of 180 rpm. all experiments were carried out with ferrous ion concentration of 20gl–1 , the process was studied as a function of incubation time and media PH. A set of nine 250 ml erlenmeyer flasks were used in the range of pH: 1–3 , with an increment of 0.25 pH.the concentrations of Fe 3+ and Fe total in the flasks were daily measured. the oxidation rate of ferrous ion was also determined . results showed that the pH of cultured medis was gradually increased , but the hydrolysis of ferric iron caused pH decrease. this mixed culture was able to oxidize inorganic substances such as ferrous sulfate as non–carbon source of energy for the cell growth.<\div>
